ABSTRACT

Resumen El uso de técnicas no invasivas ni estresantes para determinar perfiles hormonales, como la medición de esteroides fecales, ha incrementado la comprensión de la fisiología reproductiva en animales silvestres. Debido a la escasa información con respecto a perfiles hormonales reproductivos del perezoso de dos dedos, Choloepus hoffmani, se realizó un estudio en hembras en cautiverio en el centro de rescate "Sloth Sanctuary" (Cahuita, Limón, Costa Rica) con el fin de determinar (i) la confiabilidad de la extracción de progesterona y estradiol en heces, y su cuantificación en el analizador AIA-360®, (ii) evaluar los parámetros sanguíneos en esta especie y (iii) establecer si existe una correlación entre los esteroides plasmáticos y fecales. El estudio se realizó en un periodo de tres meses, durante noviembre de 2013 a enero de 2014, con un total de 208 muestras de heces provenientes de cinco hembras sexualmente maduras, con peso promedio de 6.32 kg. El promedio de las concentraciones medianas en las heces de las cinco hembras fue 124.21 ng/g para progesterona y 1 708.95 pg/g de estradiol. En plasma, los valores de mediana fueron 1.26 ng/mL con un mínimo de 0.32 ng/mL y 12.84 ng/mL como valor máximo; los valores plasmáticos de estrógeno se encontraron por debajo del límite de detección del equipo (25 pg/mL). Aunque no se encontró una correlación estadísticamente significativa entre la progesterona plasmática y la fecal, nuestros datos sugieren que los eventos plasmáticos se reflejan en heces durante los dos días posteriores. Asimismo, los niveles de progesterona se mantuvieron elevados durante la primera mitad de noviembre, y posteriormente mostraron una reducción importante en todas las hembras. Nuestros resultados demuestran que las extracciones en heces y su medición en el AIA-360® permiten la detección y el seguimiento de variaciones hormonales en C. hoffmani, aunque no remplaza las mediciones plasmáticas para determinar valores absolutos.


Abstract In wild animal species, the use of non-invasive and non-stressful procedures to determine hormone profiles, such as fecal steroid measurements, has considerably increased the comprehension of their reproductive physiology. Since there is limited information related to the reproductive hormone profiles of the two-toed sloth, Choloepus hoffmani, a study was conducted in captive specimens at the "Sloth Sanctuary" (Cahuita, Limón, Costa Rica), in order to determine: (i) the reliability of the fecal progesterone and estrogen extraction and its quantification with an AIA-360® analyzer, (ii) assess blood parameters in this species and (iii) evaluate if there is a correlation between fecal and plasmatic steroids. The study was performed over a three-month period, from November, 2013 to January, 2014, with a total amount of 208 fecal samples collected from five sexually mature females weighing 6.32 kg in average. The average of the median concentrations of progesterone in feces of the five females was 124.21 ng/g, and 1 708.95 pg/g for estrogen. The average minimal and maximal values were 50.96 ng/g and 1 057.46 ng/g for progesterone and, 1 191.77 pg/g and 2 159.24 pg/g for estradiol. In plasma, progesterone median values were 1.26 ng/mL, showing a minimum of 0.32 ng/mL and 12.84 ng/mL as maximum values. The plasmatic estrogen levels were below the detection limit of the equipment (25 pg/mL). Although there was no strong statistical correlation between the fecal and plasmatic progesterone fluctuations, our data suggests that the plasmatic events are mostly reflected in feces two days afterwards. Also, the levels of progesterone were elevated during the first half of November and, subsequently, showed a successive and important reduction in all the females tested. Finally, our results demonstrated that fecal steroid extractions and their measurement in a AIA-360®, allowed the successful detection and represents an alternative non-invasive determination of hormone profiles in C. hoffmani. ABSTRACT

The aim of this research was to evaluate whether the application of two plant growth-promoting (rhizo)bacteria might reduce nitrogen fertilization doses in cotton. We used strains Azotobacter chroococcum AC1 and AC10 for their proven ability to promote seed germination and cotton growth. These microorganisms were characterized by their plant growth-promoting activities. Then, we conducted a glasshouse study to evaluate the plant growth promoting ability of these strains with reduced doses of urea fertilization in cotton. Results revealed that both strains are capable of fixing nitrogen, solubilizing phosphorus, synthesizing indole compounds and producing hydrolytic enzymes. After 12 weeks, the glasshouse experiment showed that cotton growth was positively influenced due to bacterial inoculation with respect to chemical fertilization. Notably, we observed that microbial inoculation further influenced plant biomass (p<0.05) than nitrogen content. Co-inoculation, interestingly, exhibited a greater beneficial effect on plant growth parameters compared to single inoculation. Moreover, similar results without significant statistical differences were observed among bacterial co-inoculation plus 50% urea and 100% fertilization. These findings suggest that coinoculation of A. chroococcum strains allow to reduce nitrogen fertilization doses up to 50% on cotton growth. Our results showed that inoculation with AC1 and AC10 represents a viable alternative to improve cotton growth while decreasing the N fertilizer dose and allows to alleviate the environmental deterioration related to N pollution.


El objetivo de esta investigación fue evaluar si la aplicación de 2 (rizo)bacterias promotoras del crecimiento vegetal podría reducir la dosis de fertilizante nitrogenado en el cultivo de algodón. Se usaron las cepas Azotobacter chroococcum AC1 y AC10 por su habilidad para promover la germinación de semillas y el crecimiento del algodonero. Estos microorganismos fueron caracterizados sobre la base de sus actividades de promoción del crecimiento vegetal. Luego se realizó un estudio de invernadero con plantas de algodón para evaluar la capacidad de promoción del crecimiento vegetal de dichas cepas con dosis reducidas de urea. Los resultados revelaron que ambas cepas son capaces de fijar nitrógeno, solubilizar fósforo, sintetizar compuestos indólicos y producir enzimas hidrolíticas. Después de 12 semanas, el experimento de invernadero permitió observar que el crecimiento del algodón fue influido positivamente por la inoculación bacteriana con respecto a la fertilización química. En particular, se evidenció que la inoculación microbiana impactó más en la biomasa vegetal (p<0,05) que en el contenido de nitrógeno. Curiosamente, la coinoculación exhibió un mayor efecto positivo sobre los parámetros de crecimiento en comparación con la inoculación simple. Además, se observaron resultados similares, sin diferencias estadísticamente significativas, entre la coinoculación bacteriana más del 50% de urea y el 100% de fertilización. Estos hallazgos indican que la coinoculación de las cepas de A. chroococcum AC1 y AC10 permitiría reducir las dosis de fertilización nitrogenada del cultivo de arroz en hasta el 50% y aliviar, de esta manera, el deterioro ambiental relacionado con la contaminación por N.

ABSTRACT

Microorganisms that live inside and around a plant can supply it with essential substances, such as phytohormones and essential nutrients. The present investigation aimed to isolate and characterize the phyllosphere, the endophytic, and the water tank bacteria associated with Vriesea gigantea and Tillandsia aeranthos. The bacteria were tested for siderophore and indole-3-acetic acid (IAA) production, phosphate solubilization, and presence of the nif H gene. Genetic diversity of the bacterial isolates was evaluated by rep-PCR. Sixty-eight bacterial strains were isolated from 3 different microhabitats of V. gigantea and from 2 microhabitats of T. aeranthos bromeliad plants. Gram-positive, spore-forming bacilli comprised most bacterial isolates. All isolates produced IAA in vitro in presence of very low amounts of tryptophan. More than 70% of the evaluated bacteria presented the ability of siderophore production and phosphate solubilization, and possessed the nif H gene. It was not possible to distinguish well-defined groups of isolates based on the bromeliad species and microhabitat they inhabit using genetic characterization by rep-PCR. Water tanks presented the most abundant diversity compared with phyllosphere and endophytes, probably due to the high nutrient concentration, which promotes an ideal environment for complex microbial communities.


Microrganismos que habitam o interior e a superfície podem fornecer substancias essenciais ao crescimento das plantas, como fitormônios e nutrientes essenciais. O presente trabalho teve como objetivo isolar e caracterizar as bactérias da filosfera, do ambiente endofítico e a água de tanque associadas à Vriesea gigantea e Tillandsia aeranthos. As bactérias foram submetidas a testes de verificação de produção de sideróforos e de ácido indol acético (AIA), solubilização de fosfatos, e a presença do gene nif H. A diversidade genética dos isolados bacterianos foi analisada por rep-PCR. Sessenta e oito microrganismos foram isolados de 3 microambientes distintos de V. gigantea e de 2 microambientes de T. aeranthos. A maioria das bactérias isoladas foram bacilos formadores de esporos, gram-positivos. Todos os isolados produziram AIA in vitro na presença de quantidades pequenas de triptofano. Mais de 70% das bactérias analisadas produziram sideróforos, solubilizaram fosfatos e possuíam o gene nif H. Não foi possível distinguir grupos definidos de microrganismos baseados no microhabitat e na espécie de bromélia de onde foram isolados usando rep-PCR. A água do tanque apresentou maior diversidade microbiana quando comparada com a filosfera e o ambiente endofítico, provavelmente devido à alta concentração de nutrientes, que promove um ambiente favorável para o desenvolvimento de comunidades microbianas complexas.

ABSTRACT

En esta investigación se pretende estimular el desarrollo de protocormos de Masdevallia coccinea y Maxillaria nutans bajo condiciones in vitro utilizando diferentes concentraciones de ácido indol acético y carbón activado. El protocolo obtenido es una alternativa de la conservación de orquídeas que se encuentran en vías de extinción, y además permite contribuir con el mejoramiento medioambiental. En la evaluación del desarrollo vegetativo bajo condiciones in vitro de los protocormos de Masdevallia coccinea y Maxillaria nutans, orquídeas en vías de extinción, se obtuvo que solamente Maxillaria nutans alcanzará su desarrollo exitoso mediante el cultivo in vitro en el medio Murashige y Skoog (1962), donde se evaluó el efecto de la interacción entre el carbón activado (0,0; 0,5, 1,0 % (p/v)) y el ácido indol acético (0,0; 0,5; 1,0 mg/L-1). El medio de cultivo empleado fue enriquecido con sacarosa al 3% y el Myo inositol al 0,1 g/L-1. Los protocormos de Masdevallia coccinea evidenciaron que se encontraban inmaduros, situación por la cual no lograron su desarrollarlo vegetativo en el experimento planteado, en atención a que se encontraban en el periodo de latencia. Por esta razón, es de gran importancia tener en cuenta que las cápsulas de las orquídeas deben estar bien maduras, a fin de garantizar que los protocormos que se forman in vitro sean maduros y completen con facilidad su morfofisiológía. Mientras que los protocormos de Maxillaria nutans dieron un mayor rendimiento en su desarrollo vegetativo. En esta investigación se determinó que el efecto de la interacción de 0,5% de carbón activado con 0,5 mg/L-1 de AIA es positivo sobre la tasa de crecimiento para el desarrollo de los protocormos de Maxillaria nutans bajo condiciones in vitro.


The evaluation of protocorms growth of Masdevallia coccinea and Maxillaria nutans under in vitro conditions, endangered orchids, it got only Maxillaria nutans reached its successful development by culture in vitro in Murashige and Skoog (1962), which evaluated the effect of the interaction between the activated carbon (0.0, 0.5, 1,0% (w / v)) and indole acetic acid (0.0, 0.5, 1.0 mg.L-1). The culture medium used was enriched with 3% sucrose and 0.1 g/L-1 Myo inositol. Protocorms of Masdevallia coccinea, showed that they were immature, state why they did not achieve their vegetative development in the experiment raised, considering that they were in the latency period. It is therefore very important to note that the capsules of the orchid should be ripe, to ensure that protocorms formed in vitro are mature and they respond with ease morphphysiology development. While protocorms of Maxillaria nutans, gave a higher yield in its vegetative development. This research found that the interaction effect of 0.5% activated carbon with 0.5 mg/L-1 of IAA is positive on the growth rate for the development of Maxillaria nutans protocorms under in vitro conditions. The protocol obtained in this investigation is an alternative of conservation of orchids that are endangered and can contribute to further environmental improvement.

El desarrollo vegetativo de los protocormos de Epidendrum elongatum Jacq., una orquídea endémica y en vías de extinción, se logró eficazmente mediante el cultivo in vitro en el medio Murashige y Skoog (1962), donde se evaluó el efecto de la interacción entre los siguientes tres factores: carbón activado (0,0; 0,5; 1,0% (w/v)); ácido indol acético (0,0; 0,5; 1,0 mg.L-1); bencil amino purina (0,0; 0,5; 1,0 mg.L-1). El medio de cultivo empleado fue enriquecido con sacarosa al 3%, y el Myo inositol al 0,1 g.L-1. En este estudio se evaluó que el efecto sobre la tasa de crecimiento de la interacción del carbón activado en concentraciones de 0,5 y 1,0% con 0,5 mg.L-1 de AIA es positivo para el desarrollo de los protocormos de E. elongatum bajo condiciones in vitro, mientras que la interacción del BAP, tanto en concentraciones de 0,5 mg.L-1 y 1,0 mg.L-1, muestra resultados no tan favorables en cuanto al grado de desarrollo de los protocormos estudiados. Este estudio describe un protocolo que permite alcanzar más de 270.000 plántulas en excelente desarrollo vegetativo dentro de 30 semanas, a partir de una cápsula de esta importante orquídea. Este protocolo es un modelo de conservación para especies que se encuentran en vías de extinción, y además permite la propagación a gran escala de Epidendrum elongatum.


The vegetative development of protocorm-like bodies (PLB) from Epidendrum elongatum Jacq. (an endemic and endangered orchid) was effectively achieved through in vitro culture in Murashige and Skoog medium (1962); the effect of the interaction between the following three factors was studied: activated charcoal (0.0, 0.5, 1.0% (w/v)), indol acetic acid (0.0, 0.5, 1.0 mg.L-1), benzylaminopurine (0.0, 0.5, 1.0 mg.L-1). The culture medium being gerused was enriched with 3% sucrose and 0.1 g.L-1 myo-inositol. This study revealed that the effect of 0.5% and 1.0% activated charcoal concentrations interacting with 0.5 mg.L-1 AIA on PBL growth rate was positive for E. elongatum PBL development in vitro conditions, whereas the interaction of BAP (at both 0.5 mg.L-1 and 1.0 mg.L-1 concentration) did not produce such favourable results regarding the degree of development of the PLB being studied. This study describes a protocol leading to more than 270,000 seedlings enjoying excellent vegetative growth being obtained within 30 weeks from a single capsule from this important orchid. This protocol represents a model for conserving endangered species and promotes large-scale Epidendrum elongatum propagation.

ABSTRACT

OBJECTIVE:To study the effect of Qinglutongbi granules on bone destruction in rats with adjuvant-induced arthritis (AIA). METHODS:AIA model was established in rats. Qinglutongbi granules were given to the model rats on day 7 for a total of 49 days. Roentgenography of joints of rats was performed for the observation of bone destruction. Synovial cells were taken from each group for cultivation of supernatant liquid, which was then added to cartilage cells that cultured in vitro. The outcome measures were proliferative response of cartilage cells, NO content and expression of antileptic type iNOS in supernatant of cartilage cells. RESULTS:Qinglutongbi granules (high, middle and low dosages) could improve the reduced bone density of joints in hind limb, widened articular cavity space and disorganization of joints in rats. Supernatant liquid that cultivated with synovial cells from AIA rats receiving Qinglutongbi granules at dosages of 7.2 g?kg-1 and 14.4 g?kg-1 could enhance the reproductive activity of chondrocytes and reduce iNOS expression as well as NO content. CONCLUSION:At a certain dosage, Qinglutongbi granules can inhibit the destruction of cartilage and bone in rats.

ABSTRACT

Objective To observe the effects of ilexonin A (IA) on IL-6 and M-CSF following ballon angioplasty in rabbit common carotide artery to provide experimental basis for percutaneous coronary interventions. Methods 30 Japanese rabbits were fed with high cholesterol food for 4 weeks. Then they were divided into three groups randomly. Each group had ten rabbits. ①Control group: the incision was sew directly after right carotide artery of the rabbit was seeked. ②Balloon dilation group:the proximal of the carotide artery was cuted,the ballon was delivered and distended,after it was drawn repeatly,the incision was closed. ③IA therapy group: operation was the same to the balloon dilation group,then IA was administered in vein.All of them were fed with high cholesterol diet for 4 weeks and the blood samples were collected 1 d before the operation and 1 d,1,2,4 weeks after the operation. The serum IL-6 and M-CSF levels were determined with radioimmunoassay.The pathological changes of injuried artery were observed.
